Background: Procollagen C-proteinase enhancer-1 PCPE-1 ; is a glycoprotein that increases the rate of proteolytic removal of the carboxyl-propeptide from procollagen by procollagen C-proteinases. Methods: Six mAbs were purified from ascitic fluids by affinity chromtography on protein-G- and PCPE-1-Sepharose. Their ability to detect PCPE-1 was tested by ELISA, immunoblotting, immunoprecipitation and immunofluorescence microscopy. Results: All of the antibodies detected PCPE-1 in ELISA and immunoblotting. Some were also active in immunoprecipitation. mAb 7A11 5 bound to the NTR domain, apparently, near its C-terminus, thereby inhibiting both cell attachment to PCPE-1 and binding of PCPE-1 to heparin. mAb 7A11 1 bound to the CUB2 domain. Immunofluorescense studies with this Ab revealed that PCPE-1 is associated with the cell membrane and the extracellular matrix of 3T3 mouse fibroblasts and rat stromal bone marrow cells. These interactions of PCPE-1 were competed for by heparin. Conclusions: mAbs 7A11 1 and 7A11 5 are useful reagents, revealing that PCPE-1 interaction with cell-surface and matrix proteoglycans is mediated by the NTR domain and suggesting that the C-terminus of PCPE-1 is critical for this interaction. Bcl-2 and Bcl-XL are anti-apoptotic proteins that are hypothesized to bind a subset of pro-apoptotic proteins, BH3 domain only proteins such as Bid and Bim. By sequestering the BH3 domain only proteins, Bcl-2 and Bcl-XL prevent the activation of the multidomain pro-apoptotic proteins Bak and Bax and therefore inhibit apoptosis. Previously, we have shown Bcl-2 can undergo conformational change during apoptosis. Using a cysteine specific lipid impermeable agent 4-acetamido-4 - iodoacetly ; amino ; stilbene2, 2 -disulfonic acid IASD ; to monitor this change we determined that upon doxorubicin apoptosis cysteine 158 moves into the lipid bilayer where it is protected from the reagent. To determine if BH3 only proteins can trigger this change we added a peptide of the BH3 domain of Bim purified to whole cell lysates obtained through nitrogen cavitation. We then determined the conformation of Bcl-2 through IASD labeling of the Bcl-2 cysteines. Following an hour incubation with Bim, 93% of the Bcl-2 cysteine 158 becomes protected from IASD.
